DNA fragment elution

 

   1. Dialysis tube electrophoresis

 

     1) gel electrophoresis

     2) cut out gel desired region

     3) 1x TAE로 잘 씻겨낸 dialysis bag에 gel fragment를 넣는다

     4) gel fragment가 잠길 만큼 멸균된 1x TAE를 dialysis bag에 넣는다

     5) dialysis bag을 electrophoresis kit에 넣고 electrophoresis : 100V, 1hr

     6) 다시 방향을 바꾸어서 electrophoresis : 100V 5-10sec

     7) dialysis bag안의 1x TAE를 새로운 tube로 옮긴다

     a8) one volume 1-butanol extraction

     b9) additon 2-3volume 100% EtOH, 1/10 3M NaOAc, -20 30min

     10) centrifugation : 12,000rpm, 30min and washing

     11) dissolve in TE (pH 8.0)

 

    a : 1-butanol은 DNA에 binding되어 있는 EtBr을 제거시켜주고 DNA를 농축시켜준다.

    b : DNA가 protein이 많이 포함되어 있어서 지저분하다고 판단되면, EtOH down전에          phenol:CIAA extraction을 해주면 된다.
