   1. Dialysis tube electrophoresis

 

     1) gel electrophoresis

     2) cut out gel desired region

     3) 1x TAE·Î Àß ¾Ä°Ü³½ dialysis bag¿¡ gel fragment¸¦ ³Ö´Â´Ù

     4) gel fragment°¡ Àá±æ ¸¸Å­ ¸ê±ÕµÈ 1x TAE¸¦ dialysis bag¿¡ ³Ö´Â´Ù

     5) dialysis bagÀ» electrophoresis kit¿¡ ³Ö°í electrophoresis : 100V, 1hr

     6) ´Ù½Ã ¹æÇâÀ» ¹Ù²Ù¾î¼­ electrophoresis : 100V 5-10sec

     7) dialysis bag¾ÈÀÇ 1x TAE¸¦ »õ·Î¿î tube·Î ¿Å±ä´Ù

     a8) one volume 1-butanol extraction

     b9) additon 2-3volume 100% EtOH, 1/10 3M NaOAc, -20 30min

     10) centrifugation : 12,000rpm, 30min and washing

     11) dissolve in TE (pH 8.0)

 

    a : 1-butanolÀº DNA¿¡ bindingµÇ¾î ÀÖ´Â EtBrÀ» Á¦°Å½ÃÄÑÁÖ°í DNA¸¦ ³óÃà½ÃÄÑÁØ´Ù.

    b : DNA°¡ proteinÀÌ ¸¹ÀÌ Æ÷ÇԵǾî À־ ÁöÀúºÐÇÏ´Ù°í ÆÇ´ÜµÇ¸é, EtOH downÀü¿¡          phenol:CIAA extractionÀ» ÇØÁÖ¸é µÈ´Ù.
